چکیده

tau is one of several types of microtubule-associated proteins (maps), responsible for the assembly and stability of microtubule networks that is present only in neurons and predominantly localized in axons which its functions are tightly regulated by phosphorylation. via as yet unknown mechanisms, tau becomes hyperphosphorylated and accompanies with neuronal degeneration, loss of synapses, aberrant calcium homeostasis, imbalanced neurotransmitter release, and ultimately with neuronal death which aggregated in several neurodegenerative diseases, collectively known as tauopathies including alzheimer's disease (ad), several frontotemporal dementias and etc. neurodegeneration is the progressive loss of structure or function of neurons and neuroinflammation is a critical marker of these diseases, in addition to protein aggregates. this review looks at various factors that are considered for explaining mechanism of tau proteins that can induce neurodegeneration. these different factors include two conformations of phosphorylated tau (cis p-tau and trans p-tau), the level of ph-tau expression, hyper phosphorylation of certain amino acids in tau proteins, mutations in the tau gene (mapt), tau neurotoxicity and etc. taking together, these results help to consider tau as a remarkable factor for treatment of tauopathies. the effect of tau disturbances on neurodegenerative disorders has been looked in many different ways. according to these results, tau can be studied as a potential target for drug development especially in the field of ad and other treatment options including immunotherapy.

Oxidative stress mediates tau-induced neurodegeneration in Drosophila.

Markers of oxidative damage have been detected in brain tissue from patients with Alzheimer disease (AD) and other neurodegenerative disorders. These findings implicate oxidative injury in the neurodegenerative process, but whether oxidative stress is a cause or a consequence of neurotoxicity remains unclear. We used a Drosophila model of human tauopathies to investigate the role of oxidative s...

Soluble oligomers from a non-disease related protein mimic Abeta-induced tau hyperphosphorylation and neurodegeneration.

Protein aggregation and amyloid accumulation in different tissues are associated with cellular dysfunction and toxicity in important human pathologies, including Alzheimer's disease and various forms of systemic amyloidosis. Soluble oligomers formed at the early stages of protein aggregation have been increasingly recognized as the main toxic species in amyloid diseases. To gain insight into th...

Tau gene mutations and neurodegeneration.

Abundant neurofibrillary lesions made of the microtubule-associated protein tau constitute a defining neuropathological characteristic of Alzheimer's disease. Filamentous tau protein deposits are also the defining neuropathological characteristic of other neurodegenerative diseases, many of which are frontotemporal dementias or movement disorders, such as Pick's disease, progressive supranuclea...

A DNA damage-activated checkpoint kinase phosphorylates tau and enhances tau-induced neurodegeneration.

Hyperphosphorylation of the microtubule associated protein tau is detected in the brains of individuals with a range of neurodegenerative diseases including Alzheimer's disease (AD). An imbalance in phosphorylation and/or dephosphorylation of tau at disease-related sites has been suggested to initiate the abnormal metabolism and toxicity of tau in disease pathogenesis. However, the mechanisms u...

NMNAT suppresses tau-induced neurodegeneration by promoting clearance of hyperphosphorylated tau oligomers in a Drosophila model of tauopathy.

Tauopathies, including Alzheimer's disease, are a group of neurodegenerative diseases characterized by abnormal tau hyperphosphorylation that leads to formation of neurofibrillary tangles. Drosophila models of tauopathy display prominent features of the human disease including compromised lifespan, impairments of learning, memory and locomotor functions and age-dependent neurodegeneration visib...
